Don Aguirre leads Spanish conquistadors to South America in 1556 in search of the Lagrimus de Cristal  tree, whose flowers are said to lift curses, heal wounds, and cure disease. The Puka Michuna tribe used the flowers of the tree to heal sick conquistadors who narrowly escaped their death. Aguirre stabs the tribal chief and sets fire to the village when he refuses to tell where the tree is. The conquistadors are immortalized by the dying chief's curse, which prevents them from leaving sight of the Amazon River without being drawn back by the jungle.

Dr. Lily Houghton's research on the tears of the moon was presented by her brother MacGregor to the Royal Society in London in 1916. The Huttons request access to the recently obtained arrowhead artifact in an attempt to revolutionize medicine and the British war effort. However, the request is turned down as the tree is considered a myth and an insult to female scientists. Lily steals Arrowhead and his old Amazon map because she thinks they are the key to finding the tree. She is narrowly saved by Prince Joachim, who is also determined to find the Tree for Germany.

Lily and McGregor arrive in Brazil in search of a guide to help them travel the Amazon River. They hire Skipper Frank Wolfe, who offers wild cruises featuring corny puns and staged threats. He initially refuses, citing the risks posed by the river and woods, but after seeing the arrowhead, he reconsiders. The three escape on Joachim's U-boat after Frank steals the engine of a boat captured by Harbourmaster Nilo.
